Should be possible to get pretty decent tones out of humbuckers, although it probably won't sound exactly like a strat single coil. If you have coil split that should add a bit more versatility.
I don't know what guitar Nokie Edwards is playing here but it does have humbuckers. Sounds quite twangy though so might be coil split? At least his latest "hitchhiker" signature models had SD humbuckers with coil split according to some interview I saw..
Feels to me like for getting balls in a clean tone it helps to run the amp as loud as possible without distorting, and to EQ in quite a lot of mids and highs... This has the downside that most overdrives will turn the sound to a really loud unusable mess (at least for home listening). Everything is a trade-off... for example heavier gauge strings seem to improve the tone but once you end up with a wound G, bends become quite difficult. It's not even purely a strength issue, you just run out of space on the fretboard
